  During the week of 2/5/24 the group and I made 250 mL of multiple types of media for our control (E. coli) and for our samples of D. sonorensis. We made the following types of media: R2A, TSA, NB, TGY, TGY w/ salt (1%, 2%, and 3%), LB, and Mueller Hilton. Our selective and differential media were EMB, PEA, MacConkey, and Mannitol. We also made BHI but did not pour/inoculate with that. We also started the inoculation process for D. sonorensis and actually spread both the control and the samples. Chad had made the differential media along with the R2A. The recipes for the media we did make are as follows:  LB - Tryptone 10.0 g/L, Yeast extract 5.0 g/L, NaCl (salt) 10.0 g/L, Agar 15.0 g/L TGY - Tryptone 3.0 g/L, Glucose 1.0 g/L, Yeast extract 3.0 g/L, Agar 15.0 g/L TSA - TSA mix 40.0 g/L NB - NB mix 8.0 g/L, NaCl (salt) 5.0 g/L, Agar 15.0 g/L TGY 1% salt - Tryptone 3.0 g/L, Glucose 1.0 g/L, Yeast extract 3.0 g/L, Agar 15.0 g/L, NaCl (salt) 10.0 g/L  TGY 2% salt - Tryptone ...

Intro-  During the week of 1/29/24 the group put E. coli through the NanoDrop machine to test the purity of the sample. Alex’s group was also tasked with doing numerous C 1 V 1 = C 2 V 2 problems to practice for the future when we need to dilute a substance. At the end of the week I watched the inoculation of Deinococcus Aquaticus. We used the OD600 setting during the process of NanoDropping E. coli. Used 2µL of E. coli sample after blanking machine and putting sample through a vortex to separate its components. As for the C 1 V 1 = C 2 V 2 p ractice, it is quite simple algebra along with some unit conversions. You need to have 3 of the variables given and then you solve for the missing one. The inoculation of aquaticus had a large focus on the actual mechanics of spreading the sample throughout the plate. The method used was to do streaks back and forth on each quarter of the plate while pulling a final streak all of the way down to the center on the last quarter.  The OD60...
